چکیده
UNC-6/Netrin is a conserved axon guidance cue that canmediate both attraction and repulsion. We previously discovered that attractive UNC-40/DCC receptor signaling stimulates growth cone filopodial protrusion and that repulsive UNC-40–UNC-5 heterodimers inhibit filopodial protrusion in C. elegans. Here, we identify cytoplasmic signaling molecules required for UNC-6-mediated inhibition of filopodial protrusion involved in axon repulsion. We show that the Rac-like GTPases CED-10 and MIG-2, the Rac GTP exchange factor UNC-73/Trio, UNC-44/Ankyrin and UNC-33/CRMP act in inhibitory UNC-6 signaling. These molecules were required for the normal limitation of filopodial protrusion in developing growth cones and for inhibition of growth cone filopodial protrusion caused by activated MYR::UNC-40 and MYR::UNC-5 receptor signaling. Epistasis studies using activated CED-10 and MIG-2 indicated that UNC-44 and UNC33 act downstream of the Rac-like GTPases in filopodial inhibition. UNC-73, UNC-33 and UNC-44 did not affect the accumulation of fulllength UNC-5::GFP and UNC-40::GFP in growth cones, consistent with a model in which UNC-73, UNC-33 and UNC-44 influence cytoskeletal function during growth cone filopodial inhibition.

HLA-B44 polymorphisms at position 116 of the heavy chain influence TAP complex binding via an effect on peptide occupancy.
A single residue polymorphism distinguishes HLA-B*4402(D116) from HLA-B*4405(Y116), which was suggested to allow HLA-B*4405 to acquire peptides without binding to tapasin-TAP complexes. We show that HLA-B*4405 is not inherently unable to associate with tapasin-TAP complexes. Under conditions of peptide deficiency, both allotypes bound efficiently to TAP and tapasin, and furthermore, random nona...
متن کاملX-ray Monitoring of Ultraluminous X-ray Sources
X-ray monitoring observations were performed with the Swift observatory of the ultraluminous Xray sources Holmberg IX X-1, NGC 5408 X-1, and NGC 4395 X-2 and also of the nuclear X-ray source in NGC 4395. Holmberg IX X-1 remains in the hard X-ray spectral state as its flux varies by a factor of 7 up to an (isotropic) luminosity of 2.8 × 10 erg s. This behavior may suggest an unusually massive co...

Evidence for Line Broadening by Electron Scattering in the Broad Line Region of Ngc 4395
A high quality Keck spectrum of the Hα line in NGC 4395 reveals symmetric exponential wings, fv ∝ e, with σ ≃ 500 km s. The wings extend out to > ∼ 2500 km s from the line core, and down to a flux density of < ∼ 10 of the peak flux density. Numerical and analytic calculations indicate that exponential wings are expected for optically thin, isotropic, thermal electron scattering. Such scattering...
متن کاملA Low - Mass Central Black Hole in the Bulgeless Seyfert 1 Galaxy Ngc 4395
NGC 4395 is one of the least luminous and nearest known type 1 Seyfert galaxies, and it also lacks a bulge. We present a Hubble Space Telescope (HST) I-band image of its nuclear region, and Keck high-resolution (∼ 8 km s−1) echelle spectra containing the Ca II near-infrared triplet. In addition to the unresolved point source, there is a nuclear star cluster of size r ≈ 3.9 pc; the upper limit o...
